Immunofluorescence (IF)

(Immunofluorescence staining of SHOC2 in A431 cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human SHOC2 polyclonal antibody (dilution ratio 1:500) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue).Positive staining was localized to Nucleus and Cytoplasm.)

SHOC2 is a scaffold protein composed almost entirely of leucine-rich repeats (LRRs) and having an N-terminal region enriched in alternating lysine and glutamate/aspartate residues (KEKE motifs). SHOC2 acts as a positive modulator of the RAS-RAF-MEK-ERK signaling cascade by favoring stable RAF1 interaction with RAS. SHOC2 mediates RAS-promoted activation of mitogen-activated protein kinase (MAPK) signaling in response to extracellular stimuli. Germline mutations in Shoc2 are associated with human developmental disease. A recurrent activating mutation in SHOC2 (p.Ser2Gly) causes Mazzanti syndrome, a RASopathy characterized by features resembling Noonan syndrome and distinctive ectodermal abnormalities. SHOC2 knockdown could affect breast cancer cell survival, and SHOC2 upregulation may be associated with a poor prognosis in patients with breast cancer.
