SELENOK Antibody detects endogenous levels of SELENOK.
Purity/Purification
The antiserum was purified by peptide affinity chromatography using SulfoLink Coupling Resin (Thermo Fisher Scientific).
Form/Format
Liquid. Rabbit IgG in phosphate buffered saline, pH7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol.
Concentration
1mg/ml (varies by lot)
Applicable Applications for anti-SELENOK antibody
ELISA (EIA)(peptide)
Application Notes
ELISA(peptide): 1:20000-1:40000
Immunogen
A synthesized peptide derived from human SELENOK.
Conjugation
Unconjugated
Similarity
Belongs to the selenoprotein K family.
Post Translational Modifications
Cleaved by CAPN2/m-calpain in resting macrophages but not in activated macrophages. Macrophage activation up-regulates expression of the calpain inhibitor CAST/calpastatin, resulting in inhibition of CAPN2 activity (By similarity).
Subunit Structure
Interacts with DERL1, DERL2, DERL3 and SELENOS. The SELENOK-SELENOS complex interacts with VCP. Interacts with ZDHHC6.
Tissue Specificity
Highly expressed in heart.
Function
Required for Ca(2+) flux in immune cells and plays a role in T-cell proliferation and in T-cell and neutrophil migration (By similarity). Involved in endoplasmic reticulum-associated degradation (ERAD) of soluble glycosylated proteins. Required for palmitoylation and cell surface expression of CD36 and involved in macrophage uptake of low-density lipoprotein and in foam cell formation (By similarity). Together with ZDHHC6, required for palmitoylation of ITPR1 in immune cells, leading to regulate ITPR1 stability and function. Plays a role in protection of cells from ER stress-induced apoptosis. Protects cells from oxidative stress when overexpressed in cardiomyocytes.
Note
Probably mainly localized in the ER.
Preparation and Storage
Store at -20 degree C. Stable for 12 months from date of receipt.

NCBI Description

This gene encodes a selenoprotein, which contains a selenocysteine (Sec) residue at its active site. The selenocysteine is encoded by the UGA codon that normally signals translation termination. The 3' UTR of selenoprotein genes have a common stem-loop structure, the sec insertion sequence (SECIS), that is necessary for the recognition of UGA as a Sec codon rather than as a stop signal. This selenoprotein is localized to the endoplasmic reticulum and is highly expressed in the heart, where it may function as an antioxidant. [provided by RefSeq, Jul 2008]

Product Notes

The SELENOK selk (Catalog #AAA9630576) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SELENOK Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's SELENOK can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA)(peptide). ELISA(peptide): 1:20000-1:40000. Researchers should empirically determine the suitability of the SELENOK selk for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SELENOK, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
