Western Blot (WB), Flow Cytometry (FC/FACS)
Application Notes
Suitable for use in Western Blot and Flow cytometry.
Dilution: Western blot: 0.1 0.2ug/ml. The detection limit is~1ng/lane under non-reducing and reducing conditions.
Flow cytometry: 25ug/ml. Add 10il of the diluted solution to 1-2.5 x 10e5 cells in a total reaction volume not exceeding 200ul. The binding of biotinylated antibodies may be visualized by adding a secondary developing reagent such as streptavidin conjugated to a fluorochrome.
Immunogen
NS0-derived recombinant human rhPlexin D1 extracellular domain.
Hybridoma
Produced in a goat immunized with purified, NS0-derived, recombinant human Plexin D1 extracellular domain (rhPlexin D1; aa 47-1271).
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol, aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for anti-PLXND1 antibody
Plexin D1 is a type I transmembrane protein that is the prototype of the Plexin D subfamily in the Plexin family. It is expressed on multiple tissues including fetal neuronal and endothelial cells. Plexin D1 expression is also induced on tumor vasculature and tumor cells. The 1879 amino acid (aa) mature human Plexin D1 contains a large (1225 aa) extracellular domain with a 500 aa sema domain and three short MRS (Met-related sequences) cysteine-rich motifs. By alternative splicing, multiple Plexin-D1 isoforms may exist. Plexin D1 binds Sema4A to suppress

Uniprot Description

PLXND1: Cell surface receptor for SEMA4A and for class 3 semaphorins, such as SEMA3A, SEMA3C and SEMA3E. Plays an important role in cell-cell signaling, and in regulating the migration of a wide spectrum of cell types. Regulates the migration of thymocytes in the medulla. Regulates endothelial cell migration. Plays an important role in ensuring the specificity of synapse formation. Required for normal development of the heart and vasculature. Mediates anti-angiogenic signaling in response to SEMA3E. Belongs to the plexin family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Receptor, misc.

Chromosomal Location of Human Ortholog: 3q22.1

Cellular Component: integral to plasma membrane; lamellipodium; plasma membrane; intracellular

Molecular Function: protein domain specific binding; protein binding; semaphorin receptor activity

Biological Process: patterning of blood vessels; axon guidance; synaptogenesis; positive regulation of protein binding; endothelial cell migration; angiogenesis; regulation of cell migration; regulation of angiogenesis

Product Notes

The PLXND1 plxnd1 (Catalog #AAA616656) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Plexin D1 (Biotin) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Plexin D1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Flow Cytometry (FC/FACS). Researchers should empirically determine the suitability of the PLXND1 plxnd1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Plexin D1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
