Western Blot (WB) (Figure 1. Western blot analysis of PAI1 using anti-PAI1 antibody (MBS1750452). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: rat small intestine tissue lysates,Lane 2: rat kidney tissue lysates,Lane 3: mouse kidney tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PAI1 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for PAI1 at approximately 45-50KD. The expected band size for PAI1 is at 45KD. )

Related Product Information for anti-PAI1 antibody
Plasminogen activator inhibitor-1 (PAI-1) also known as endothelial plasminogen activator inhibitoror serpin E1 is a protein that in humans is encoded by the SERPINE1 gene. This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.

NCBI Description

mediates inhibition of fibrinolysis by inhibiting the activity of plasminogen activator; may promote neuronal survival [RGD, Feb 2006]

Uniprot Description

Serine protease inhibitor. Inhibits TMPRSS7. Is a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As PLAT inhibitor, it is required for fibrinolysis down-regulation and is responsible for the controlled degradation of blood clots. As PLAU inhibitor, it is involved in the regulation of cell adhesion and spreading. Acts as a regulator of cell migration, independently of its role as protease inhibitor. It is required for stimulation of keratinocyte migration during cutaneous injury repair. It is involved in cellular and replicative senescence (). Plays a role in alveolar type 2 cells senescence in the lung (). Is involved in the regulation of cementogenic differentiation of periodontal ligament stem cells, and regulates odontoblast differentiation and dentin formation during odontogenesis ().

Product Notes

The PAI1 serpine1 (Catalog #AAA1750452) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PAI1 Picoband antibody reacts with Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's PAI1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Direct ELISA. WB: 0.1-0.5 ug/ml Direct ELISA: 0.1-0.5 ug/ml Tested Species: In-house tested species with positive results. Other applications have not been tested. Optimal dilutions should be determined by end users. Enhanced Chemiluminescent Kit with anti-Rabbit IgG MBS176460 for western blot. Researchers should empirically determine the suitability of the PAI1 serpine1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PAI1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
