Intended Uses: This rat plasminogen activator inhibitor type 1 (PAI-1) total antigen assay is intended for the quantitative determination of total PAI-1 in biological fluids.

Principle of the Assay: Pat PAI-1 will bind to the capture antibody coated on the microtiter plate. Free, latent, and complexed PAI-1 will will react with the capture antibody on the plate. Any unbound PAI-1 is washed away and an anti PAI-1 primary antibody is added. Excess primary antibody is washed away and bound antibody, which is proportional to the total PAI-1 present in the samples, is then reacted with a secondary antibody. Following an additional washing step, TMB is then used for color development at 450nm. The amount of color development is directly proportional to the concentration of total PAI-1 in the sample.

Background: Plasminogen activator inhibitor-1 (PAI-1) is a central regulator of the blood fibrinolytic system [1]. Clinical studies have indicated that increased PAI-1 levels increase the risk for thrombosis, whereas decreased levels may cause recurrent bleeding [2].

NCBI Description

mediates inhibition of fibrinolysis by inhibiting the activity of plasminogen activator; may promote neuronal survival [RGD, Feb 2006]

Uniprot Description

SERPINE1: a secreted protein that acts as 'bait' for tissue plasminogen activator, urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. Belongs to the serpin family. Interacts with VTN. Binds LRP1B; binding is followed by internalization and degradation. Plasma levels of PAI-1 and VCAM-1 together may be useful in predicting post-operative recurrence in patients with colorectal cancer.

Product Notes

The General SERPINE1 serpine1 (Catalog #AAA135541) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA135541 ELISA Kit recognizes General SERPINE1. It is sometimes possible for the material contained within the vial of "PAI-1 total antigen assay, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
