Nuclear factor kappa B (NFkB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NFkB mediates the expression of a great variety of genes in response to extracellular stimuli including IL-1, TNF alpha LPS and mitogens. A serine/threonine protein kinase which mediates NFkappa;B activation by IL-1, TNF alpha; and CD95 was identified recently and designated NIK (for NF kappa;B inducing kinase). NIK is an activator of I kappa B kinase alpha; and beta; (IKK alpha; and IKK beta;) (2-5). Therefore, NIK is a key molecule in the NF kappa;B signaling pathway leading to the induction of a variety of gene expression in response to proinflammatory cytokines and bacteria products.

NCBI Description

This gene encodes mitogen-activated protein kinase kinase kinase 14, which is a serine/threonine protein-kinase. This kinase binds to TRAF2 and stimulates NF-kappaB activity. It shares sequence similarity with several other MAPKK kinases. It participates in an NF-kappaB-inducing signalling cascade common to receptors of the tumour-necrosis/nerve-growth factor (TNF/NGF) family and to the interleukin-1 type-I receptor. Function: Lymphotoxin beta-activated kinase which seems to be exclusively involved in the activation of NF-kappa-B and its transcriptional activity. Promotes proteolytic processing of NFKB2/P100, which leads to activation of NF-kappa-B via the non-canonical pathway. Could act in a receptor-selective manner. Ref.8

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Subunit structure: Interacts with TRAF2, TRAF5, TRAF6, IKKA and NFKB2/P100

Post-translational modification: Autophosphorylated. Phosphorylation at Thr-559 is required to activates its kinase activity and 'Lys-63'-linked polyubiquitination. Phosphorylated by CHUK/IKKA leading to MAP3K14 destabilization. Ref.11 Ref.15Ubiquitinated. Undergoes both 'Lys-48'- and 'Lys-63'-linked polyubiquitination. 'Lys-48'-linked polyubiquitination leads to its degradation by the proteasome, while 'Lys-63'-linked polyubiquitination stabilizes and activates it. Ref.8 Ref.15
