Western Blot (WB)

(Detection of Human RelA/NFKB p65 by Western Blot. Western blot shows lysates of K562 human chronic myelogenous leukemia cell line, Daudi human Burkitt's lymphoma cell line, and LNCaP human prostate cancer cell line. PVDF membrane was probed with 1ug/ml of Human RelA/NFkB p65 Antigen Affinity purified Polyclonal Antibody followed by HRP-conjugated Sheep IgG Secondary Antibody. A specific band was detected for RelA/NFKB p65 at approximately 70kD. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

Immunoprecipitation (IP)

(Detection of RelA/NFKB p65-regulated Genes by Chromatin Immunoprecipitation. Jurkat human acute T cell leukemia cell line treated with 50ng/ml PMA and 200ng/ml calcium ionomycin for overnight was fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. RelA/NFkB p65/DNA complexes were immunoprecipitated using 5ug Sheep Human RelA/NFkB p65 Antigen Affinity­purified Polyclonal Antibody or control antibody for 15min in an ultrasonic bath, followed by Biotinylated Sheep IgG Secondary Antibody. Immunocomplexes were captured using 50ul of MagCellect Streptavidin Ferrofluid and DNA was purified using chelating resin solution. The p21 promoter was detected by standard PCR.)

RelA belongs to a family of transcription factors (NFKB) that play a fundamental role in inflammatory and immune responses. The NFKB complex is composed of a heterodimer of a Rel family member (RelA, c-Rel, RelB) and either NFKB1 or NFKB2 subunits. RelA and NFKB1 are the most common heterodimeric pair. The NFKB complex is sequestered in the cytoplasm by inhibitory IKB proteins. Upon cellular activation, the ubiquitin-proteosome pathway degrades the IKB proteins allowing the NFKB complex to translocate to the nucleus and activate gene transcription.

NKIRAS2: Atypical Ras-like protein that acts as a potent regulator of NF-kappa-B activity by preventing the degradation of NF-kappa-B inhibitor beta (NFKBIB) by most signals, explaining why NFKBIB is more resistant to degradation. May act by blocking phosphorylation of NFKBIB and nuclear localization of p65/RELA NF- kappa-B subunit. It is unclear whether it acts as a GTPase. Both GTP- and GDP-bound forms block phosphorylation of NFKBIB. Belongs to the small GTPase superfamily. Ras family. KappaB-Ras subfamily. 3 isoforms of the human protein are produced by alternative splicing.
