Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NENF is a secreted protein that is expressed in neurons but not glial cells of the brain. This protein has neurotrophic activity in primary cultured mouse neurons but not mitogenic activity in primary cultured mouse astrocytes. NENF activated mitogen-activated protein (MAP) and phosphotidylinositol-3 kinase pathways and could be inhibited by pertussis toxin but not receptor tyrosine kinases, suggesting that NENF acts through a Gi/Go-protein-coupled receptor. Despite its predicted molecular weight of ~20kDa, NENF is observed at 37kDa in SDS-PAGE.

This gene encodes a neurotrophic factor that may play a role in neuron differentiation and development. A pseudogene of this gene is found on chromosome 12. Alternate splicing of this gene results in multiple transcript variants. [provided by RefSeq, Jan 2009]

NENF: Displays neurotrophic activity and activates phosphorylation of MAPK1/ERK2, MAPK3/ERK1 and AKT1/AKT in primary cultured neurons. Does not have mitogenic activity in primary cultured astrocytes. May play a role on neuronal differentiation and may have a transient effect on neural cell proliferation in neural precursor cells. Neurotrophic activity is enhanced by binding to heme. Belongs to the cytochrome b5 family. MAPR subfamily.
