NENF is a secreted protein that is expressed in neurons but not glial cells of the brain. This protein has neurotrophic activity in primary cultured mouse neurons but not mitogenic activity in primary cultured mouse astrocytes. NENF activated mitogen-activated protein (MAP) and phosphotidylinositol-3 kinase pathways and could be inhibited by pertussis toxin but not receptor tyrosine kinases, suggesting that NENF acts through a Gi/Go-protein-coupled receptor. Despite its predicted molecular weight of ~20kD, NENF is observed at 37kD in SDS-PAGE.

Uniprot Description

Function: Displays neurotrophic activity and activates phosphorylation of MAPK1/ERK2, MAPK3/ERK1 and AKT1/AKT in primary cultured neurons. Does not have mitogenic activity in primary cultured astrocytes. May play a role on neuronal differentiation and may have a transient effect on neural cell proliferation in neural precursor cells. Neurotrophic activity is enhanced by binding to heme. Ref.1 Ref.4 Ref.5

Tissue specificity: In the embryo, expressed most abundantly in brain and spinal cord. Widely expressed in adult tissues including brain, heart, lung and kidney. In brain, expressed in neurons but not in glial cells. Ref.1

Developmental stage: In embryonic cerebral cortex is first weakly detected at E12.5, and thereafter gradually increased. At E13.5 expression is observed mostly in the preplate. Ref.4

Domain: The cytochrome b5 heme-binding domain was proven to bind heme, although it lacks the conserved iron-binding His residue at position 81.
