Rabbit anti-Human Nestin Polyclonal Antibody | anti-NES antibody
Nestin Antibody (S1409)
Western Blot (WB)
(Anti-Nestin Antibody (S1409)at 1:2000 dilution + Hela whole cell lysatesLysates/proteins at 20 ug per lane.SecondaryGoat Anti-Rabbit IgG, (H+L), Peroxidase conjugated at 1/10000 dilutionPredicted band size : 177 kDaBlocking/Dilution buffer: 5% NFDM/TBST.)
Immunofluorescence (IF)
(Fluorescent confocal image of SY5Y cells stained with Nestin (S1409) antibody. SY5Y cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with MBS9203158 Nestin (S1409) primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min). Note the highly specific localization of the Nestin (S1409) immunosignal to the intermediate filaments, supported by Human Protein Atlas Data (http://www.proteinatlas.org/ENSG00000132688).)
Western Blot (WB)
(Western blot analysis of Nestin Antibody (S1409) in HepG2 cell line lysates (35ug/lane). NES (arrow) was detected using the purified Pab.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human hepatocarcinoma reacted with Nestin Antibody (S1409), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.)
Flow Cytometry (FC/FACS)
(Flow cytometric analysis of HepG2 cells using Nestin Antibody (S1409)(bottom histogram) compared to a negative control cell (top histogram). FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)

NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of the intermediate filament protein family and is expressed primarily in nerve cells. [provided by RefSeq, Sep 2011]
Uniprot Description
nestin: an intermediate filament protein. It is expressed predominantly in stem cells of the central nervous system in the neural tube. Upon terminal neural differentiation, nestin is downregulated and replaced by neurofilaments.
Protein type: Cytoskeletal
Chromosomal Location of Human Ortholog: 1q23.1
Cellular Component: intermediate filament cytoskeleton; cytoplasm; intermediate filament
Molecular Function: intermediate filament binding; structural molecule activity
Biological Process: positive regulation of intermediate filament depolymerization; central nervous system development; negative regulation of catalytic activity; brain development; negative regulation of neuron apoptosis; cell projection morphogenesis; negative regulation of protein binding; G2/M transition of mitotic cell cycle; embryonic camera-type eye development

Product Notes
The NES nes (Catalog #AAA9203158) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 1389-1416. The Nestin Antibody (S1409) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Nestin can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Immunohistochemistry (IHC), Immunofluorescence (IF), Flow Cytometry (FC/FACS). WB~~1:1000. Researchers should empirically determine the suitability of the NES nes for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Nestin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
