Specificity
This item specifically recognises 49kDa MEK5, also known as MAP2K5, when phosphorylated at Ser311 and Thr315.

MEK5 is a dual specificity serine/threonine protein kinase belonging to the MAP kinase kinase family, which acts as a key regulator in a mitogen-activated protein (MAP) kinase cascade, and hence growth factor-induced cellular stimulation. MEK5 exists in naturally occurring MEK5alpha and MEK5beta splice variants, the former of which is known to specifically activate/regulate MAPK7/ERK5/BMK1, whilst the latter may act as a suppressor of MAPK7 activation. Activation of MEK5 itself is regulated by upstream MAP3K3/MEKK3, and phosphorylation of MEK5 at both the Ser311 and Thr315 sites has been implicated in cell proliferation.

Applicable Applications for anti-MEK 5 antibody
Western Blot (WB)*
Application Notes
Western Blot: This item detects a band of approximately 49kDa in rat testes cell lysates.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
Perservative Stabilisers
0.09% Sodium Azide (NaN3)
0.01% Bovine Serum Albumin
50% Glycerol
Immunogen
Synthetic phosphopeptide corresponding to an amino acid sequence within MEK5, which includes phoshorylated Ser311 and Thr315.
Buffer Solution
Antiserum Preparation
Antisera to MEK5 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Target Species
Rat
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.

kinase involved in signal transduction in the cell [RGD, Feb 2006]

Uniprot Description

MEK5: a dual specificity protein kinase of the STE7 family. Activates ERK5 and is itself phosphorylated and activated by MEKK3, as well as by atypical protein kinase C isoforms. Involved in growth factor stimulated cell proliferation and muscle cell differentiation. Four alternatively spliced isoforms have been described.

Product Notes

The MEK 5 map2k5 (Catalog #AAA222783) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI RAT MEK 5 (pSer311/Thr315) reacts with Bovine (Expected from Sequence), Chicken (Expected from Sequence), Dog (Expected from Sequence), Human (Expected from Sequence), Mouse (Expected from Sequence), Primate (Expected from Sequence), Xenopus (Expected from Sequence), Zebrafish (Expected from S and may cross-react with other species as described in the data sheet. AAA Biotech's MEK 5 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB)*. Researchers should empirically determine the suitability of the MEK 5 map2k5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MEK 5, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
