Related Product Information for anti-MAPKK5 (MEK5) (pS311/pT315) antibody
The MEK5 (also known as MKK5, MAPKK5, SKK5) is a dual specificity serine/threonine protein kinase belonging to the MAP kinase kinase family. It catalyzes tyrosine phosphorylation of ERK5 whereas MEK5 itself is regulated by MEKK3. An important link between MEK5 and metastatic prostrate cancer has been demonstrated. Dual phosphorylation of Ser311 and Thr315 have been implicated in cell proliferation.

NCBI Description

The protein encoded by this gene is a dual specificity protein kinase that belongs to the MAP kinase kinase family. This kinase specifically interacts with and activates MAPK7/ERK5. This kinase itself can be phosphorylated and activated by MAP3K3/MEKK3, as well as by atypical protein kinase C isoforms (aPKCs). The signal cascade mediated by this kinase is involved in growth factor stimulated cell proliferation and muscle cell differentiation. Three alternatively spliced transcript variants of this gene encoding distinct isoforms have been described. [provided by RefSeq]

Uniprot Description

Function: Acts as a scaffold for the formation of a ternary MAP3K2/MAP3K3-MAP3K5-MAPK7 signaling complex. Activation of this pathway appear to play a critical role in protecting cells from stress-induced apopotosis, neuronal survival and cardiac development and angiogenesis. Ref.1 Ref.2

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Cofactor: Magnesium.

Subunit structure: Interacts with PARD6A, MAP3K3 and MAPK7. Forms a complex with SQSTM1 and PRKCZ or PRKCI

By similarity. Interacts with Yersinia yopJ. Ref.1 Ref.7

Tissue specificity: Expressed in many adult tissues. Abundant in heart and skeletal muscle.

Domain: Binds MAP3K2/MAP3K3 and MAPK7 via non-overlapping residues of the OPR domain. This domain also mediates interactions with SQSTM1 and PARD6A

Post-translational modification: Activated by phosphorylation on Ser/Thr by MAP kinase kinase kinases

By similarity.Yersinia yopJ may acetylate Ser/Thr residues, preventing phosphorylation and activation, thus blocking the MAPK signaling pathway.

Sequence similarities: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase subfamily.Contains 1 OPR domain.Contains 1 protein kinase domain.

Product Notes

The MAPKK5 (MEK5) (pS311/pT315) map2k5 (Catalog #AAA462077) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit anti MEK5 (pSer311/ pThr315, Phospho-specific) reacts with Human, Rat, Mouse, Chicken, Canine and may cross-react with other species as described in the data sheet. AAA Biotech's MAPKK5 (MEK5) (pS311/pT315) can be used in a range of immunoassay formats including, but not limited to, ELISA, Western Blot. Western Blot: 0.1-1 ug/ml ELISA: 0.01-0.1 ug/ml Immunoprecipitation: 2-5 ug/ml Immunohistochemistry: 2-5 ug/ml. Researchers should empirically determine the suitability of the MAPKK5 (MEK5) (pS311/pT315) map2k5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MAPKK5 (MEK5) (pS311/pT315), Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
