The JNK (c-Jun NH2-terminal kinase) signal transduction cascade is one of three identified MAP kinase pathways in mammals. JNK pathway activation occurs in response to environmental stress, is associated with the nuclear accumulation of JNK itself, and is implicated in the immune response, cellular proliferation, oncogenic transformation, morphogenesis, and apoptosis. JIP1 (JNK-interacting protein-1, also known as Islet Brain-1 and mitogen-activated protein kinase 8-interacting protein-1,) is a cytoplasmic protein that possesses an N-terminal JNK-binding domain and a C-terminal SRC homology (SH3) domain. Its SH3 domain is related to the SH3 domains of the tyrosine kinase c-Fyn and the p85 subunit of phosphoinositide-3 kinase (PI3K). Overexpression of JIP1 causes cytoplasmic retention of JNK, thereby preventing its nuclear accumulation and the subsequent activation of the JNK pathway. JIP1: a member of the JNK-interacting protein group of scaffold proteins that selectively mediates JNK signaling by aggregating specific components of the MAPK cascade. Required for JNK activation in response to excitotoxic stress. Causes inhibition of JNK-regulated activity by retaining JNK in the cytoplasm and by inhibiting the JNK phosphorylation of c-Jun. May also participate in ApoER2-specific reelin signaling. Directly or indirectly regulates GLUT2 gene expression and beta-cell function.
