Application Notes
Applications Validated:
WB: 1:500-1:2,000 dilution
Note: Many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt/Sonication Protocol when preparing nuclear extracts for Western blot.

Preparation and Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20 degree C for up to 2 years. Keep all reagents on ice when not in storage.1ug/ul

Related Product Information for anti-H3T3ph antibody
Short Description: Our Histone H3T3ph antibody (pAb) was raised in a Rabbit host. It has been validated for use in Dot blot, Immunocytochemistry, Immunofluorescence and Western blot; it has been shown to react with Human samples, but it is predicted that it will react with a wide range of sample types.

Background: Histone H3 is one of the core components of the nucleosome. The nucleosome is the smallest subunit of chromatin and consists of 147 base pairs of DNA wrapped around an octamer of core histone proteins (two each of Histone H2A, Histone H2B, Histone H3 and Histone H4). Chromatin is subject to a variety of chemical modifications, including post-translational modifications of the histone proteins and the methylation of cytosine residues in the DNA. Reported histone modifications include acetylation, methylation, phosphorylation, ubiquitylation, glycosylation, ADP-ribosylation, carbonylation and SUMOylation; these modifications play a major role in regulating gene expression. In vivo, phosphorylation of histone H3 at threonine 3 begins at early prophase in the vicinity of the nuclear envelope, spreads to the pericentromeric chromatin during prometaphase and is fully reversed by late anaphase, during mitosis.

Product Notes

The H3T3ph (Catalog #AAA388387) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Histone H3T3ph antibody (pAb) reacts with Human, Wide Range Predicted and may cross-react with other species as described in the data sheet. AAA Biotech's Histone H3T3ph can be used in a range of immunoassay formats including, but not limited to, Dot Blot (DB), Immunocytochemistry (ICC), Immunofluorescence (IF), Western Blot (WB). Applications Validated: WB: 1:500-1:2,000 dilution Note: Many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt/Sonication Protocol when preparing nuclear extracts for Western blot. Researchers should empirically determine the suitability of the H3T3ph for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Histone H3T3ph, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
