Rabbit Histone H3K14me1 Polyclonal Antibody | anti-H3K14me1 antibody
Histone H3K14me1 antibody (pAb)
WB: 1:500-1:2,500 dilution
Note: Many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt/Sonication Protocol when preparing nuclear extracts for Western blot.
Western Blot (WB)
(Histone H3K14me1 antibody (pAb) tested by Western blot. Nuclear extract of HeLa cells (20 ug) probed with H3K14me1 antibody at a 1:500 dilution.)
Dot Blot (DB)
(Specificity Data: Dot blot analysis was used to confirm the specificity of Histone H3K14me1 antibody (pAb) for monomethyl-lysine 14 of histone H3. Peptides corresponding to regions around major sites of histone H3 methylation were spotted onto PVDF and probed with the antibody at a dilution of 1:30,000. The amount of peptide (in picomoles) spotted is indicated next to each row.Top panel - Lane 1: unmodified Lys14. Lane 2: monomethyl Lys14. Lane 3: dimethyl Lys14. Lane 4: trimethyl Lys14. Immunofluorescence (IF)
(Detection of H3K4me1 by immunofluorescence U2OS cells were stained with H3K4me1 antibody at a dilution of 1:500. Left panel: DAPI. Middle panel: H3K4me1 antibody staining. Right panel: merge.)
Background: Histone H3 is one of the core components of the nucleosome. The nucleosome is the smallest subunit of chromatin and consists of 147 base pairs of DNA wrapped around an octamer of core histone proteins (two each of Histone H2A, Histone H2B, Histone H3 and Histone H4). Chromatin is subject to a variety of chemical modifications, including post-translational modifications of the histone proteins and the methylation of cytosine residues in the DNA. Reported histone modifications include acetylation, methylation, phosphorylation, ubiquitylation, glycosylation, ADP-ribosylation, carbonylation and SUMOylation; these modifications play a major role in regulating gene expression. Histone methylation can be associated with transcriptional activation or repression, depending on the methylated residue.

Product Notes
The H3K14me1 (Catalog #AAA388929) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Histone H3K14me1 antibody (pAb) reacts with Human, Wide Range Predicted and may cross-react with other species as described in the data sheet. AAA Biotech's Histone H3K14me1 can be used in a range of immunoassay formats including, but not limited to, Dot Blot (DB), Immunofluorescence (IF), Western Blot (WB). Applications Validated: WB: 1:500-1:2,500 dilution Note: Many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt/Sonication Protocol when preparing nuclear extracts for Western blot. Researchers should empirically determine the suitability of the H3K14me1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Histone H3K14me1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
