Western Blot (WB) (Western blot analysis of FOXO1 was performed by loading 25 ug of HeLa (lane 1), 293T (lane 2) and rat heart lysates (lane 3) onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4 degree C overnight. The membrane was probed with FOXO1 polyclonal antibody at a dilution of 1/500 overnight at 4 degree C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Detection was performed using chemiluminescence. Images were taken at an exposure time of 2 min (lanes 1 and 2) and 1 min (lane 3). Results show a band at ~66 kDa)

Preperation
Antiserum to FOXO1 was raised by repeated immunization of rabbits with highly purified antigen.
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred.
Storage in frost-free freezers is not recommended. This product should be stored undiluted.
Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. Shelf Life: 12 months from date of despatch.

Rabbit anti FOXO1 antibody recognizes Forkhead box protein O1, also known as FOXO1, Forkhead box protein O1A or Forkhead in rhabdomyosarcoma.

FOXO1 like other members of the forkhead protein family acts as a transcription factor by binding to DNA with its forkhead box protein domain. The subcellular activation and thereby the transcriptional activity of FOXO1 is regulated via AKT mediated phosphorylation while its levels are regulated via polyubiquitination.

FOXO1 has been shown to play a role in various signaling pathways ranging from apoptosis, proliferation, oxidative stress, DNA damage to insulin signaling (Huang and Tindall 2007). The protein has also been proposed to act as a tumor suppressor (Fu and Tindall 2008, Xie et al..2012).

This gene belongs to the forkhead family of transcription factors which are characterized by a distinct forkhead domain. The specific function of this gene has not yet been determined; however, it may play a role in myogenic growth and differentiation. Translocation of this gene with PAX3 has been associated with alveolar rhabdomyosarcoma. [provided by RefSeq, Jul 2008]
