IHC: 1:50-1:200
ELISA(peptide): 1:20,000-1:40,000
Western Blot (WB)
(Western blot analysis of extracts from Mouse brain, using EB3 Antibody. The lane on the left was treated with blocking peptide.)
Immunohistochemistry (IHC)-Paraffin
(MBS9612457 at 1/100 staining Rat brain tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612457 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612457 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Subcellular Location: Cytoplasm>Cytoskeleton. Note: Associated with the microtubule network. Detected at the plus end of microtubules.
Tissue Specificity: Predominantly expressed in brain and muscle.
Subunit Structure: Interacts with APC2 (PubMed:10644998). Homodimer (PubMed:19255245). Heterodimer with MAPRE1 (PubMed:19255245). Interacts with DCTN1 and SRCIN1 (PubMed:14514668, PubMed:19146815). Binds to the C-terminal domain of APC (PubMed:14514668). Binds monomeric and polymerized tubulin (PubMed:10188731). Interacts (via C-terminus) with CLIP1 (PubMed:17563362). Interacts with SLAIN2 and SLAIN1 (PubMed:21646404). Interacts with AKAP9 (PubMed:28814570). Interacts with PDE4DIP (PubMed:28814570). Interacts with PDE4DIP isoform 13/MMG8/SMYLE; this interaction is required for its recruitment to the Golgi apparatus (PubMed:25217626).
Similarity: Composed of two functionally independent domains. The N-terminal domain forms a hydrophobic cleft involved in microtubule binding and the C-terminal is involved in the formation of mutually exclusive complexes with APC and DCTN1.Belongs to the MAPRE family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)32kDa.
NCBI Description
The protein encoded by this gene is a member of the RP/EB family of genes. The protein localizes to the cytoplasmic microtubule network and binds APCL, a homolog of the adenomatous polyposis coli tumor suppressor gene. [provided by RefSeq, Jul 2008]
Uniprot Description
EB3: Binds to the plus end of microtubules and regulates the dynamics of the microtubule cytoskeleton. Promotes microtubule growth. May be involved in spindle function by stabilizing microtubules and anchoring them at centrosomes. May play a role in cell migration. Belongs to the MAPRE family. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: Microtubule-binding
Chromosomal Location of Human Ortholog: 2p23.3-p23.1
Cellular Component: microtubule cytoskeleton; cytoplasmic microtubule; perinuclear region of cytoplasm; cytoplasm; midbody
Molecular Function: protein binding; microtubule binding
Biological Process: mitosis; positive regulation of cyclin-dependent protein kinase activity; cell division; positive regulation of transcription, DNA-dependent

Product Notes
The MAPRE3 mapre3 (Catalog #AAA9612457) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The EB3 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Sheep(100%), Rabbit(100%), Dog(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's EB3 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). IF: 1:100-1:500 ICC: 1:100-1:500 WB: 1:500-1:2000 IHC: 1:50-1:200 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the MAPRE3 mapre3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "EB3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
