Western Blot (WB)
(Western blot analysis of CPB2 expression in human placenta extract (lane 1) and HEPG2 whole cell lysates (lane 2). CPB2 at 48KD, 60KD was detected using rabbit anti- CPB2 Antigen Affinity purified polyclonal antibody at 0.5 ug/mL. The blot was developed using chemiluminescence (ECL) method. )
Immunohistochemistry (IHC)
(CPB2 was detected in paraffin-embedded sections of human lung cancer tissues using rabbit anti- CPB2 Antigen Affinity purified polyclonal antibody at 1ug/mL. The immunohistochemical section was developed using SABC method. )
Background: Carboxypeptidase B2 (CPB2), also known as carboxypeptidase U (CPU), plasma carboxypeptidase B (pCPB) or thrombin-activatable fibrinolysis inhibitor (TAFI), is an enzyme that, in humans, is encoded by the gene CPB2. CPB2 is synthesized by the liver and circulates in the plasma as a plasminogen-bound zymogen. When it is activated by proteolysis at residue Arg92 by the thrombin/thrombomodulin complex, CPB2 exhibits carboxypeptidase activity. Activated CPB2 reduces fibrinolysis by removing the fibrin C-terminal residues that are important for the binding and activation of plasminogen.

NCBI and Uniprot Product Information
NCBI Description
Carboxypeptidases are enzymes that hydrolyze C-terminal peptide bonds. The carboxypeptidase family includes metallo-, serine, and cysteine carboxypeptidases. According to their substrate specificity, these enzymes are referred to as carboxypeptidase A (cleaving aliphatic residues) or carboxypeptidase B (cleaving basic amino residues). The protein encoded by this gene is activated by trypsin and acts on carboxypeptidase B substrates. After thrombin activation, the mature protein downregulates fibrinolysis. Polymorphisms have been described for this gene and its promoter region. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Jun 2013]
Uniprot Description
CPB2: Cleaves C-terminal arginine or lysine residues from biologically active peptides such as kinins or anaphylatoxins in the circulation thereby regulating their activities. Down- regulates fibrinolysis by removing C-terminal lysine residues from fibrin that has already been partially degraded by plasmin. Belongs to the peptidase M14 family. 2 isoforms of the human protein are produced by alternative splicing.

The Anti-CPB2 Antibody reacts with Human No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's CPB2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Paraffin, ELISA (EIA). Western Blot: 0.1-0.5mug/ml; Tested Species: Human Immunohistochemisry Paraffin: 0.5-1mug/ml; Tested Species: Human ELISA: 0.1-0.5mug/ml; Tested Species: Human Tested Species:In-house tested species with positive results. By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections. Other applications have not been tested. Researchers should empirically determine the suitability of the CPB2 cpb2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CPB2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
