Application Notes
Suitable for use in Western Blot, ELISA and Neutralization.
Dilution: Western Blot: 1-5ug/ml using ECL.
ELISA: 0.5-2.0ug antibody and 10-50ng of control antigen/well.
Neutralization: 100-200ug IgG neutralizes 50% activity of 1ug of Angiogenin. Neutralizes biological activity of recombinant human Angiogenin as measured by measurement of ribonucleolytic activity using tRNA.
Optimal dilution determined by the researcher.

Human Angiogenin protein expressed in E. coli.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Embryonic vascular system undergoes a series of complex, highly regulated series of events involving differentiation, migration and association of primitive endothelial cells. This process is termed vasculogenesis. A further remodeling of the primitive vascular system forms the mature cardiovascular system. This process is known as angiogenesis (sprouting of new capillary vessels from pre-existing vasculature). Angiogenesis accounts for the formation of vasculature into previously avascular organs such as brain and kidney. Angiogenic activity in the adult is required during the normal tissue repair, and for the remodeling of the female reproductive organs (ovulation and placental development). Certain pathological conditions, such as tumor growth and diabetic retinopathy, also require angiogenesis. Angiogenin is a single polypeptide chain of 123aa (mol wt ~14kD) secreted by tumor cells. It is a potent inhibitor of neovascularization. It specifically binds to endothelial cells and elicits second messenger systems. It has ribonucleolytic activity and is 33% identical to pancreatic RNAse A.

ANG: May function as a tRNA-specific ribonuclease that abolishes protein synthesis by specifically hydrolyzing cellular tRNAs. Binds to actin on the surface of endothelial cells; once bound, angiogenin is endocytosed and translocated to the nucleus. Angiogenin induces vascularization of normal and malignant tissues. Angiogenic activity is regulated by interaction with RNH1 in vivo. Defects in ANG are the cause of susceptibility to amyotrophic lateral sclerosis type 9 (ALS9). ALS is a degenerative disorder of motor neurons in the cortex, brain stem and spinal cord. ALS is characterized by muscular weakness and atrophy. Belongs to the pancreatic ribonuclease family.
