Related Product Information for anti-A2ML1 antibody
A2ML1 is a new member of the alpha 2-macroglobulin protease inhibitor family, and shares most the characteristics of this family. It is the first family member detected in the epidermis and may play an important role during desquamation by inhibiting extracellular proteases.

NCBI Description

This gene encodes a member of the alpha-macroglobulin superfamily. The encoded protein acts as an inhibitor for several proteases, and has been reported as the p170 antigen recognized by autoantibodies in the autoimmune disease paraneoplastic pemphigus (PNP; PMID: 20805888). Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2012]

Uniprot Description

Function: Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase

By similarity. Displays inhibitory activity against chymotrypsin, papain, thermolysin, subtilisin A and, to a lesser extent, elastase but not trypsin. May play an important role during desquamation by inhibiting extracellular proteases. Ref.5

Subunit structure: Monomer. Ref.5

Subcellular location: Secreted Ref.5.

Tissue specificity: In the epidermis, expressed predominantly in the granular layer at the apical edge of keratinocytes (at protein level). Also detected in placenta, testis and thymus but not in epithelia of kidney, lung, small intestine or colon. Ref.5

Developmental stage: Up-regulated during keratinocyte differentiation. Ref.5

Sequence similarities: Belongs to the protease inhibitor I39 (alpha-2-macroglobulin) family. [View classification]

Sequence caution: The sequence BAB71612.1 differs from that shown. Reason: Erroneous initiation. The sequence BAC04793.1 differs from that shown. Reason: Erroneous initiation. The sequence BAC85653.1 differs from that shown. Reason: Erroneous initiation. The sequence BAC85654.1 differs from that shown. Reason: Erroneous initiation.

Product Notes

The A2ML1 a2ml1 (Catalog #AAA648921) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The A2ML1, NT (A2ML1, CPAMD9, Alpha-2-macroglobulin-like protein 1, C3 and PZP-like alpha-2-macroglobulin domain-containing protein 9) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's A2ML1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Flow Cytometry (FC/FACS). Suitable for use in Western Blot, Flow Cytometry, ELISA Dilution: ELISA: 1:1,000 Western Blot: 1:100-500 Flow Cytometry: 1:10-50. Researchers should empirically determine the suitability of the A2ML1 a2ml1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "A2ML1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
