Intended Uses: Rat tPA total antigen assay is intended for the quantitative determination of total tissue plasminogen activator in rat plasma and other biological fluids.

Principle of the Assay: Rat tPA will bind to the capture antibody coated on the microtiter plate. Free, latent, and complexed enzyme will react with the capture antibody on the plate. A standard calibration curve is prepared using dilutions of tPA along with the samples to be measured. After appropriate washing steps, monoclonal anti-rat tPA primary antibody binds to the captured enzyme. Excess antibody is washed away and bound monoclonal antibody is then reacted with the secondary antibody conjugated to horseradish peroxidase. TMB substrate is used for color development at 450nm. The amount of color development is directly proportional to the concentration of tPA antigen in the sample.

Background: Tissue plasminogen activator (tPA) is a serine protease that converts plasminogen to the active serine protease plasmin in the blood fibrinolytic system [1,2]. It also plays an important role in the removal of incipient thrombi[3]. tPA is widely used for the thrombolytic treatment of acute myocardial infarction [3].

NCBI Description

serine protease; catalyzes conversion of inactive proenzyme plasminogen into active serine protease plasmin [RGD, Feb 2006]

Uniprot Description

Function: Converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling and degradation, in cell migration and many other physiopathological events.

Catalytic activity: Specific cleavage of Arg-|-Val bond in plasminogen to form plasmin.

Enzyme regulation: Inhibited by SERPINA5

By similarity.

Subunit structure: Heterodimer of chain A and chain B held by a disulfide bond. Binds to fibrin with high affinity. This interaction leads to an increase in the catalytic efficiency of the enzyme due to an increase in affinity for plasminogen. Similarly, binding to heparin increases the activation of plasminogen. Binds to annexin A2, cytokeratin-8, fibronectin and laminin. Binds to mannose receptor and the low-density lipoprotein receptor-related protein (LRP1); these proteins are involved in TPA clearance. Binds LRP1B; binding is followed by internalization and degradation. Forms heterodimer with SERPINA5

By similarity.

Subcellular location: Secreted › extracellular space.

Domain: Both FN1 and one of the kringle domains are required for binding to fibrin

By similarity. Both FN1 and EGF-like domains are important for binding to LRP1

By similarity. The FN1 domain mediates binding to annexin A2

By similarity. The second kringle domain is implicated in binding to cytokeratin-8 and to the endothelial cell surface binding site

By similarity.

Post-translational modification: The single chain, almost fully active enzyme, can be further processed into a two-chain fully active form by a cleavage after Arg-308 catalyzed by plasmin, tissue kallikrein or factor Xa.

Sequence similarities: Belongs to the peptidase S1 family.Contains 1 EGF-like domain.Contains 1 fibronectin type-I domain.Contains 2 kringle domains.Contains 1 peptidase S1 domain.

Product Notes

The General PLAT plat (Catalog #AAA135505) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA135505 ELISA Kit recognizes General PLAT. It is sometimes possible for the material contained within the vial of "tPA total antigen assay, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
