Related Product Information for VCAM-1 elisa kit
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-VCAM-1 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-VCAM-1 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the VCAM-1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of VCAM-1 can be calculated.

Background: Vascular cell adhesion protein 1 (VCAM-1), also known as cluster of differentiation 106 (CD106) is a protein that in humans is encoded by the VCAM1 gene. This gene contains 9 exons spanning about 25 kb, and It is expressed on both large and small blood vessels only after the endothelial cells are stimulated by cytokines. The VCAM-1 is a cell surface sialoglycoprotein, a type I membrane protein that is a member of the Ig superfamily. It mediates the adhesion of lymphocytes, monocytes, eosinophils, and basophils to vascular endothelium. It also functions in leukocyte-endothelial cell signal transduction, and it may play a role in the development of atherosclerosis and rheumatoid arthritis.

NCBI Description

binds integrin very late antigen (VLA4); plays a role in cell adhesion [RGD, Feb 2006]

Uniprot Description

Function: Important in cell-cell recognition. Appears to function in leukocyte-endothelial cell adhesion. Interacts with integrin alpha-4/beta-1 (ITGA4/ITGB1) on leukocytes, and mediates both adhesion and signal transduction. The VCAM1/ITGA4/ITGB1 interaction may play a pathophysiologic role both in immune responses and in leukocyte emigration to sites of inflammation.

Subunit structure: Binds to ECMV-D capsid proteins and acts as a receptor for this virus

By similarity.

Subcellular location: Membrane; Single-pass type I membrane protein.

Tissue specificity: Expressed on inflamed vascular endothelium, as well as on macrophage-like and dendritic cell types in both normal and inflamed tissue.

Sequence similarities: Contains 7 Ig-like C2-type (immunoglobulin-like) domains.

Product Notes

The Mouse VCAM-1 vcam1 (Catalog #AAA355350) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355350 ELISA Kit recognizes Mouse VCAM-1. AAA Biotech's VCAM-1 can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of VCAM-1 in mouse serum, plasma, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the VCAM-1 vcam1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "VCAM-1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
