Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- HMG-CoA Reductase antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-HMG-CoA Reductase antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the HMG-CoA Reductase amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of HMG-CoA Reductase can be calculated.

Uniprot Description

HMGCR: the rate-limiting enzyme for cholesterol synthesis. Regulated via a negative feedback mechanism mediated by sterols and non-sterol metabolites derived from mevalonate, the product of the reaction catalyzed by this reductase. Normally in mammalian cells this enzyme is suppressed by cholesterol derived from the internalization and degradation of low density lipoprotein (LDL) via the LDL receptor. Competitive inhibitors of the reductase induce the expression of LDL receptors in the liver, which in turn increases the catabolism of plasma LDL and lowers the plasma concentration of cholesterol, an important determinant of atherosclerosis.

Product Notes

The Mouse Hmgcr hmgcr (Catalog #AAA7606267) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7606267 ELISA Kit recognizes Mouse Hmgcr. It is sometimes possible for the material contained within the vial of "HMG-CoA Reductase (3-hydroxy-3-methylglutaryl-coenzyme A reductase), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
