Intended Uses: This HMGCR ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey HMGCR. This ELISA kit for research use only!

Principle of the Assay: HMGCR ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-HMGCR antibody and an HMGCR-HRP conjugate. The assay sample and buffer are incubated together with HMGCR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HMGCR concentration since HMGCR from samples and HMGCR-HRP conjugate compete for the anti-HMGCR antibody binding site. Since the number of sites is limited, as more sites are occupied by HMGCR from the sample, fewer sites are left to bind HMGCR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HMGCR concentration in each sample is interpolated from this standard curve.

Uniprot Description

HMGCR: the rate-limiting enzyme for cholesterol synthesis. Regulated via a negative feedback mechanism mediated by sterols and non-sterol metabolites derived from mevalonate, the product of the reaction catalyzed by this reductase. Normally in mammalian cells this enzyme is suppressed by cholesterol derived from the internalization and degradation of low density lipoprotein (LDL) via the LDL receptor. Competitive inhibitors of the reductase induce the expression of LDL receptors in the liver, which in turn increases the catabolism of plasma LDL and lowers the plasma concentration of cholesterol, an important determinant of atherosclerosis.
