Principle of the Assay: The Picokine Mouse Fabp2 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Mouse Fabp2 with a 96-well strip plate that is pre-coated with antibody specific for Fabp2. The detection antibody is a biotinylated antibody specific for Fabp2. The capture antibody is a monoclonal antibody from rat, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Mouse Fabp2 with immunogen: Expression system for standard: E.coli, Immunogen sequence: A2-E132. The kit is analytically validated with ready to use reagents. To measure Mouse Fabp2, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Mouse Fabp2 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Fabp2 in the sample!!Background/Introduction: Fatty acid-binding protein 2 (FABP2), also known as Intestinal-type fatty acid-binding protein (I-FABP) is a protein that in humans is encoded by the FABP2 gene. The intracellular fatty acid-binding proteins (FABPs) belong to a multigene family with nearly twenty identified members. FABPs are divided into at least three distinct types, namely the hepatic-, intestinal-and cardiac-type. Using a human cDNA probe for in situ hybridization studies, they regionalized the assignment to 4q28-q31. The intestinal form of FABP was mapped to mouse chromosome 3 between the amylase and alcohol dehydrogenase-3 loci. Also, they may be responsible in the modulation of cell growth and proliferation.

FABP4: a lipid transport protein that is the predominant fatty acid binding protein found in adipocytes. Also expressed in macrophages. Binds both long chain fatty acids and retinoic acid. Delivers long-chain fatty acids and retinoic acid to their cognate receptors in the nucleus. Homodimer. Forms a beta-barrel structure that accommodates hydrophobic ligands in its interior. Interacts with PPARG. Monomer. FABP4 knockout mice fed a high-fat and high-calorie diet become obese but develop neither insulin resistance nor diabetes, suggesting that this protein might be a link between obesity and insulin resistance and diabetes. Mice deficient in both FABP4 and ApoE show protection against atherosclerosis when compared with mice deficient only in ApoE. Mice carrying a FABP4 genetic variant exhibit both reduced FABP4 expression and a reduced potential for developing type 2 diabetes and coronary heart disease. A related study in humans indicated a similar pattern, suggesting that FABP4 may be a potential therapeutic target in the treatment of these disorders.
