Principle of the Assay: The Mouse FABP4 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse FABP4 in Cell Culture Supernates, Serum, Plasma, Cell Lysates. This assay employs an antibody specific for Mouse FABP4 coated on a 96-well plate. Standards and samples are pipetted into the wells and FABP4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse FABP4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of FABP4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: FABP4, also known as aP2 (adipocyte Protein 2), encodes the fatty acid binding protein found in adipocytes. The FABP4 gene is mapped to 8q21. FABP4 functions as a cytosolic lipid chaperone in macrophages and is involved in regulating macrophage endoplasmic reticulum (ER) stress. Aditionally, FABP4 protein is expressed in HBEs and is strongly upregulated by both IL4 and IL13 and downregulated by IFN-gamma, suggesting a role in allergic airway inflammation.

Uniprot Description

FABP4: a lipid transport protein that is the predominant fatty acid binding protein found in adipocytes. Also expressed in macrophages. Binds both long chain fatty acids and retinoic acid. Delivers long-chain fatty acids and retinoic acid to their cognate receptors in the nucleus. Homodimer. Forms a beta-barrel structure that accommodates hydrophobic ligands in its interior. Interacts with PPARG. Monomer. FABP4 knockout mice fed a high-fat and high-calorie diet become obese but develop neither insulin resistance nor diabetes, suggesting that this protein might be a link between obesity and insulin resistance and diabetes. Mice deficient in both FABP4 and ApoE show protection against atherosclerosis when compared with mice deficient only in ApoE. Mice carrying a FABP4 genetic variant exhibit both reduced FABP4 expression and a reduced potential for developing type 2 diabetes and coronary heart disease. A related study in humans indicated a similar pattern, suggesting that FABP4 may be a potential therapeutic target in the treatment of these disorders. Belongs to the calycin superfamily,fatty-acid binding protein (FABP) family.

Protein type: Lipid-binding

Cellular Component: cytoplasm; cytosol

Biological Process: brown fat cell differentiation; cholesterol homeostasis; cytokine production; fatty acid metabolic process; negative regulation of protein kinase activity; negative regulation of transcription, DNA-dependent; positive regulation of cell proliferation; positive regulation of inflammatory response; white fat cell differentiation

Product Notes

The Mouse FABP4 fabp4 (Catalog #AAA8248434) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8248434 ELISA Kit recognizes Mouse FABP4. It is sometimes possible for the material contained within the vial of "FABP4, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
