Principle of the Assay: The Picokine Monkey THBS1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Monkey THBS1 with a 96-well strip plate that is pre-coated with antibody specific for THBS1. The detection antibody is a biotinylated antibody specific for THBS1. The capture antibody is monoclonal antibody from mouse, the detection antibody is polyclonal antibody from goat. The kit contains recombinant Monkey THBS1 with immunogen: Expression system for standard: NSO; Immunogen sequence: N19-P1170. The kit is analytically validated with ready to use reagents. To measure Monkey THBS1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Monkey THBS1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Monkey THBS1 in the sample!!Background/Introduction: Thrombospondin 1, also known as THBS1, is a protein that in humans in encoded by the THBS1 gene. Thrombospondin 1 is a subunit of a disulfide-linked homotrimeric protein. This protein is an adhesive glycoprotein that mediates cell-to-cell and cell-to-matrix interactions. By in situ hybridization, the THBS1 gene was mapped to monkey primate 15q15 and the cognate gene to monkey primate chromosome 2(region F) and was localized to 15q11-qter by Southern analysis of human-rodent somatic cell hybrids. Thrombospondin I is a multimodular secreted protein that associates with the extracellular matrix and possesses a variety of biologic functions, including a potent antiangiogenic activity. Other thrombospondin genes include thrombospondins II, III, and IV.

NCBI Description

This gene encodes a secreted ligand of the TGF-beta (transforming growth factor-beta) superfamily of proteins. Ligands of this family bind various TGF-beta receptors leading to recruitment and activation of SMAD family transcription factors that regulate gene expression. The encoded preproprotein is proteolytically processed to generate the mature protein, which binds to the activin receptor-like kinase 1 (ALK1) and plays important roles in cardiovascular development including cardiomyocyte proliferation and regulation of heart size, closure of the ductus arteriosus, angiogenesis and ventricular trabeculation.
