Principle of the Assay||The Mouse ANGPTL3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse ANGPTL3 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse ANGPTL3 coated on a 96-well plate. Standards and samples are pipetted into the wells and ANGPTL3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse ANGPTL3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ANGPTL3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: ANGPTL3, also known as Angiopoietin-like 3 or HYPLl, is a protein that in humans is encoded by the ANGPTL3 gene. It is mapped to 1p31.3. The protein encoded by this gene is a member of the angiopoietin-like family of secreted factors. It is expressed predominantly in the liver, and has the characteristic structure of angiopoietins, consisting of a signal peptide, N-terminal coiled-coil domain, and the C-terminal fibrinogen (FBN)-like domain. ANGPTL3 also acts as dual inhibitor of lipoprotein lipase (LPL) and endothelial lipase (EL), and increases plasma triglyceride and HDL cholesterol in rodents. It can inhibit endothelial lipase to catalyze HDL-phospholipid and increase HDL-PL levels.

NCBI Description

This gene encodes a member of the angiopoietin-like family of proteins. The encoded preproprotein is proteolytically processed to generate multiple protein products, which may inhibit triglyceride metabolism. Homozygous knockout mice for this gene exhibit reduced plasma lipid concentrations, including reduced plasma triglyceride concentrations, and enhanced activity of enzymes involved in triglyceride metabolism. ANGPTL3: Defects in ANGPTL3 are the cause of familial hypobetalipoproteinemia type 2 (FHBL2); also called combined hypobetalipoproteinemia familial. FHBL2 is a disorder of lipid metabolism characterized by less than 5th percentile age- and sex-specific levels of low density lipoproteins, and dietary fat malabsorption. Affected individuals present with combined hypolipidemia, consisting of extremely low plasma levels of LDL cholesterol, HDL cholesterol, and triglycerides.
