Western Blot: 1:5,000-1:10,000 detects nitrated a- or b-Synuclein recombinant protein. Nitrated and unnitrated a-synuclein recombinant protein were resolved by electrophoresis, transferred to nitrocellulose and probed with antinitro-a/b-Synuclein (1:5,000 dilution). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. nitrated a- Synuclein (~14.5kD). Note. The detected band at ~30kD represents the formation of o,o' -dityrosine cross-links occurring during nitration with peroxynitrite (5)
