Western Blot: 1:5,000-1:10,000 detects nitrated a- or b-Synuclein recombinant protein. Nitrated and unnitrated a-synuclein recombinant protein were resolved by electrophoresis, transferred to nitrocellulose and probed with antinitro-a/b-Synuclein (1:5,000 dilution). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. nitrated a- Synuclein (~14.5kD). Note. Full-length nitrated recombinant human a-synuclein.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
