Application Notes
IHC: fresh rat tissue 1:400; fresh human tissue 1:20. Fix in acetone for 5 min at 4 deg, paraformaldehyde, paraffin. Pretreat paraffin tissues in microwave for 2 x 5 min at 750 W.
ELISA: Use as a capture antibody.
Biological Significance
Surfactant protein D is acalcium dependent carbohydrate binding protein. The function of SP-D is not fully identified butit is known that SP-D enhances the production of oxygen radicals in alveolar macrophages and regulates some actions of SP-A. It has a similar structure to other C-type mammalian lectins and a molecular weightof 620 kDa (non-reducing conditions) and 43 kDa (reducing conditions).
Cross-Reactivity
Reacts with alveolar type II cells, alveolar macrophages, types of Clara cells, SP-A+ and SP-A-, and extracellular deposits. Alveolar type III cells are negative. No reaction on rat skin, colon, kidney or liver. SP-A cross reacitivity is typically evident at very low dilutions.
Immunogen
Surfactant Protein D antibody was raised using purified rat SP-D from bronchoalveolar lavage
Preparation and Storage
Store lyophilized material at 2-8 degree for up to 1 year. Reconstitute with 500 ul sterile distilled water, aliquot and store at -20 degree for up to 6 months, avoiding repeated freeze/thaw cycles.
