Principles and Procedures
Visualization of the antigen present in tissue sections is accomplished in a multi-step immunohistochemical staining process, in conjunction with a horseradish peroxidase (HRP) or alkaline phosphatase (AP) linked detection system. The process involves the addition of the stated antibody (primary antibody) to a tissue slide, followed by a secondary antibody (linked to an enzyme complex) which specifically binds to the primary antibody. A chromogenic substrate is then added which reacts with the enzyme complex, resulting in a colorimetric reaction at the site of the antigen. Results are interpreted using a light microscope.
Reconstitution
The prediluted antibody does not require any mixing, dilution, reconstitution, or titration; the antibody is ready-to-use and optimized for staining.
The concentrated antibody requires dilution in the optimized buffer, to the recommended working dilution range.
Specimen Collection and Preparation for Analysis
Each tissue section should be fixed with 10% neutral buffered formalin, cut to the applicable thickness (4um), and placed on a glass slide that is positively charged. The prepared slide may then be baked for a minimum of 30 minutes in a 53-65 degree C oven (do not exceed 24 hours).
Note: Performance evaluation has been shown on human tissues only. Variable results may occur due to extended fixation time or special processes of specific tissue preparations.
Preparation and Storage
Store at 2-8 degree C. Do not freeze.
To ensure proper stability and delivery of the antibody after each run, replace the cap and immediately place the bottle in a refrigerator in an upright position.
Positive and negative controls should be simultaneously run with unknown specimens.

Related Product Information for anti-MSH2 antibody
Intended Use: The MSH2 [IHC410] antibody is intended for qualified laboratories to qualitatively identify by light microscopy the presence of associated antigens in sections of formalin-fixed, paraffin-embedded tissue sections using IHC test methods.
Description: MutS Homolog 2 (MSH2) is a protein involved in the mismatch-repair pathway. This protein is commonly associated with hereditary non-polyposis colorectal cancer, and mutations in this gene are correlated with the development of sporadic colorectal carcinoma. Expression levels of MSH2 are abnormally low in a high percentage of patients with microsatellite instability, as well as endometrial and ovarian cancers. Use of Anti-MSH2 is optimized when paired in an IHC panel with antibodies against MSH6, MLH1, and PMS2. Reports have shown Anti-MSH2 to be useful in the detection of the protein in a number of normal and neoplastic tissues, and for identifying a loss of MSH2 in tumors that are microsatellite-unstable.

Product Notes

The MSH2 msh2 (Catalog #AAA344032) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The MSH2 reacts with Human and may cross-react with other species as described in the data sheet. The recommended working dilution range is 1:100 - 1:200. Researchers should empirically determine the suitability of the MSH2 msh2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MSH2, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
