Tonsil, Follicular Lymphoma, Diffuse Large B-Cell Lymphoma
Principles and Procedures
Visualization of the antigen present in tissue sections is accomplished in a multi-step immunohistochemical staining process, in conjunction with a horseradish peroxidase (HRP) or alkaline phosphatase (AP) linked detection system. The process involves the addition of the stated antibody (primary antibody) to a tissue slide, followed by a secondary antibody (linked to an enzyme complex) which specifically binds to the primary antibody. A chromogenic substrate is then added which reacts with the enzyme complex, resulting in a colorimetric reaction at the site of the antigen. Results are interpreted using a light microscope.
Reconstitution
The prediluted antibody does not require any mixing, dilution, reconstitution, or titration; the antibody is ready-to-use and optimized for staining.
The concentrated antibody requires dilution in the optimized buffer, to the recommended working dilution range.
Specimen Collection and Preparation for Analysis
Each tissue section should be fixed with 10% neutral buffered formalin, cut to the applicable thickness (4um), and placed on a glass slide that is positively charged. The prepared slide may then be baked for a minimum of 30 minutes in a 53-65 degree C oven (do not exceed 24 hours).
Note: Performance evaluation has been shown on human tissues only. Variable results may occur due to extended fixation time or special processes of specific tissue preparations.
Preparation and Storage
Store at 2-8 degree C. Do not freeze.
To ensure proper stability and delivery of the antibody after each run, replace the cap and immediately place the bottle in a refrigerator in an upright position.
Positive and negative controls should be simultaneously run with unknown specimens.

Related Product Information for anti-LMO2 antibody
Intended Use: The LMO2 [IHC615] antibody is intended for qualified laboratories to qualitatively identify by light microscopy the presence of associated antigens in sections of formalin-fixed, paraffin-embedded tissue sections using IHC test methods.
Description: LMO2, also known as LIM-Only transcription factor 2, RBTN2, or TTG2, is an oncoprotein that is expressed in normal germinal center B-cells, as well as bone marrow hematopoietic precursors and endothelial cells. LMO2 plays a role in angiogenesis and hematopoesis, and its expression has been detected in erythroid and myeloid precursors, megakaryocytes, and also in lymphoblastic and acute myeloid leukemias. LMO2 protein expression has been noted in diffuse large B-cell lymphoma, the most common adult non-Hodgkin lymphoma, as well as follicular lymphoma, a neoplasm derived from germinal center B-cells that accounts for a number of cases of non-Hodgkin lymphomas.

NCBI Description

LMO2 encodes a cysteine-rich, two LIM-domain protein that is required for yolk sac erythropoiesis. The LMO2 protein has a central and crucial role in hematopoietic development and is highly conserved. The LMO2 transcription start site is located approximately 25 kb downstream from the 11p13 T-cell translocation cluster (11p13 ttc), where a number T-cell acute lymphoblastic leukemia-specific translocations occur. Alternative splicing results in multiple transcript variants encoding different isoforms.[provided by RefSeq, Nov 2008]

Uniprot Description

Acts with TAL1/SCL to regulate red blood cell development. Also acts with LDB1 to maintain erythroid precursors in an immature state.

Product Notes

The LMO2 lmo2 (Catalog #AAA344063) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The LMO2 reacts with Human and may cross-react with other species as described in the data sheet. The recommended working dilution range is 1:100 - 1:200. Researchers should empirically determine the suitability of the LMO2 lmo2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "LMO2, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
