Rabbit anti-Human GABAR Monoclonal Antibody | anti-GABAR antibody
Anti-GABAR [1F4]
Immunofluorescence (IF)
(Immunofluorescence staining of fixed U251 cells with anti-GABAR antibody 1F4 (MBS488686) Immunofluorescence analysis of paraformaldehyde fixed U251 cells on Shi-fix coverslips stained with the chimeric rabbit IgG version of 1F4 () at 10ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom , DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (MBS488140) followed by staining with Alexa Fluor 488 secondary antibody.)
This antibody was raised against alpha1beta2gamma2 GABA-A receptor.
Note on publication: This article describes the generation of 1F4 antibody as well as Fab fragments derived from it for the pupose of puryfying and analysing the structure of alpha1beta2gamma2 GABA-A receptor.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a gamma-aminobutyric acid (GABA) receptor. GABA is the major inhibitory neurotransmitter in the mammalian brain where it acts at GABA-A receptors, which are ligand-gated chloride channels. Chloride conductance of these channels can be modulated by agents such as benzodiazepines that bind to the GABA-A receptor. GABA-A receptors are pentameric, consisting of proteins from several subunit classes: alpha, beta, gamma, delta and rho. Mutations in this gene cause juvenile myoclonic epilepsy and childhood absence epilepsy type 4. Multiple transcript variants encoding the same protein have been identified for this gene. Uniprot Description
GABRA1: GABA, the major inhibitory neurotransmitter in the vertebrate brain, mediates neuronal inhibition by binding to the GABA/benzodiazepine receptor and opening an integral chloride channel. Defects in GABRA1 are the cause of childhood absence epilepsy type 4 (ECA4). A subtype of idiopathic generalized epilepsy characterized by onset at age 6-7 years, frequent absence seizures (several per day) and bilateral, synchronous, symmetric 3-Hz spike waves on EEG. During adolescence, tonic-clonic and myoclonic seizures may develop. Absence seizures may either remit or persist into adulthood. Defects in GABRA1 are the cause of juvenile myoclonic epilepsy type 5 (EJM5). A subtype of idiopathic generalized epilepsy. Patients have afebrile seizures only, with onset in adolescence (rather than in childhood) and myoclonic jerks which usually occur after awakening and are triggered by sleep deprivation and fatigue. Belongs to the ligand-gated ion channel (TC 1.A.9) family. Gamma-aminobutyric acid receptor (TC 1.A.9.5) subfamily. GABRA1 sub-subfamily.

Product Notes
The GABAR gabra1 (Catalog #AAA488686) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-GABAR [1F4] reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's GABAR can be used in a range of immunoassay formats including, but not limited to, Fluorescence-Detection Size-exclusion Chromatography (FSEC), Crystallography. FSEC: High affinity and specificity of the 1F4 antibody to alpha1beta2gamma2 GABA-A receptor was demonstrated using fluorescence-detection size exclusion chromatography with GFP-tagged receptor (shift in elution volume) (Zhu et al., 2018). Crystallography: Furthermore, this antibody was utilised in production and purification of the alpha1beta2gamma2 GABA-A receptor (Zhu et al., 2018). Finally, Fab fragments derived from the antibody were used to facilitate crystallisation and improve stabilisation of the alpha1beta2gamma2 GABA-A receptor (Zhu et al., 2018). Researchers should empirically determine the suitability of the GABAR gabra1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GABAR, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
