Suitable for use in Western Blot.
Dilution: Western Blot: 1:1000. Detects a band at ~130kD for the nonglycosylated form of Flt3 and the glycosylated mature form is identified at ~160kD.
Immunogen
Synthetic phosphopeptide corresponding to the sequences surrounding Tyr591 of human Flt3.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

FMS-related tyrosine kinase 3 (FLT3, also called Flk2), a member of the type III receptor tyrosine kinase family that includes c-Kit, PDGF and M-CSF receptors, is expressed on early hematopoietic progenitor cells, and supports growth and differentiation within the hematopoietic system (1,2). FLT3 is activated after binding with its ligand FL, which results in a cascade of tyrosine autophosphorylation and tyrosine phosphorylation of downstream cellular targets (3). The p85 subunit of PI3 kinase, SHP2, GRB2 and Shc are associated with FLT3 after FL stimulation (4-6). Tyrosine 591 is located in the juxtamembrane region of FLT3. It may play an important role in regulation of FLT3 tyrosine kinase activity. Somatic mutations of FLT3 consisting of internal tandem duplications (ITDs) occur in 20% of patients with acute myeloid leukemia (7).

NCBI Description

This gene encodes a class III receptor tyrosine kinase that regulates hematopoiesis. The receptor consists of an extracellular domain composed of five immunoglobulin-like domains, one transmembrane region, and a cytoplasmic kinase domain split into two parts by a kinase-insert domain. The receptor is activated by binding of the fms-related tyrosine kinase 3 ligand to the extracellular domain, which induces homodimer formation in the plasma membrane leading to autophosphorylation of the receptor. The activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow. Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia. [provided by RefSeq, Jul 2008]

FLT3 -/- mice have an impaired developmental capacity of primitive hematopoietic progenitor cells of all lineages with the greatest impact on lymphopoietic precursors. Activating mutations found in one third of cases of acute myeloid leukemia (AML), as well as in acute lymphoblastic leukemia, acute promyelocytic leukemia and myelodysplastic syndrome.
