Assay Conditions
ADP-Glo® assay was performed with 0.2mg/ml Abltide as a substrate and 100mM ATP at final concentration. Reaction was done for 45 minutes at 30ºC. ADP-Glo® for 45 minutes at RT and Kinase detection for 55 minutes at RT.

Preparation and Storage
Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product , centrifuge the original vial after thawing and prior to removing the cap.

FMS-related tyrosine kinase 3 (FLT3, also called Flk2), a member of the type III receptor tyrosine kinase family that includes c-Kit, PDGF and M-CSF receptors, is expressed on early hematopoietic progenitor cells, and supports growth and differentiation within the hematopoietic system. FLT3 is activated after binding with its ligand FL, which results in a cascade of tyrosine autophosphorylation and tyrosine phosphorylation of downstream cellular targets. The p85 subunit of PI3 kinase, SHP2, GRB2 and Shc are associated with FLT3 after FL stimulation. Tyrosine 591 is located in the juxtamembrane region of FLT3. It may play an important role in regulation of FLT3 tyrosine kinase activity. Somatic mutations of FLT3 consisting of internal tandem duplications (ITDs) occur in 20% of patients with acute myeloid leukemia.
