Applicable Applications for anti-CD8 alpha antibody
Flow Cytometry (FC/FACS), ELISA (EIA)
Application Notes
1:50 (or use 1.6ul of concentrated stock per 5 x 105-cell test)
Performance
Reagent was tested for binding to ACK lysed murine splenocytes in FACS. Five x 105 splenocytes per tube were washed and pre incubated with 20 ul of 300 ug/ml Mouse IgG (to block non specific binding), after which they were incubated 45 minutes on ice with 80 ul of anti-Mouse CD8/APC at a 1:50 dilution (2 ug/ml). They were then washed three times, fixed and analyzed by FACS. A 9% sub population of the cells stained positive with a mean shift of 2.59 log10 fluorescent units when compared to background. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-mouse CD8 antibody.
Conjugation
APC
Production
Protein G purified antibody from tissue culture supernatant was conjugated to R-Phycoerythrin through a sulfo-ester linkage. Unconjugated antibody was removed using size exclusion chromatography. Antibody conjugate is at 0.1 mg/ml with an A650/A280 ratio of 2.12.
Buffer
50 mM Sodium Phosphate pH 7.5, 500 mM Potassium Chloride, 150mM NaCl, 15% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).
Immunogen
murine thymocytes/splenocytes
Preparation and Storage
Store at 2 - 5 degree C. Do not freeze! Protect from light.
Product should retain activity for at least 12 months after shipping date when stored as recommended.

Product Notes

The CD8 alpha (Catalog #AAA666688) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The anti-mouse CD8 alpha -APC reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CD8 alpha can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), ELISA (EIA). 1:50 (or use 1.6ul of concentrated stock per 5 x 105-cell test). Researchers should empirically determine the suitability of the CD8 alpha for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD8 alpha, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
