PE-Cy7 is excited by 488 laser. Its Emax is 785nm.
Performance
Reagent was tested for binding to ACK lysed murine splenocytes in FACS. Five x 105 splenocytes per tube were washed and pre incubated with 20 ul of 300 ug/ml Mouse IgG (to block non specific binding), after which they were incubated 45 minutes on ice with 80 ul of anti-Mouse CD4/PE-Cy7 at a 1:50 dilution (5 ug/ml). They were then washed three times, fixed and analyzed by FACS. A 21% sub population of the cells stained positive with a mean shift of 1.2 log10 fluorescent units when compared to background. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-mouse CD4 antibody.

Production
Protein G purified antibody from tissue culture supernatant was conjugated to Cy7-linked R-Phycoerythrin through a sulfo-ester linkage. Unconjugated antibody was removed using size exclusion chromatography. Antibody conjugate is at 250 ug/ml.

50 mM Sodium Phosphate pH 7.5, 500 mM Potassium Chloride, 150mM NaCl, 15% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).

Information: Antibody GK1.5 binds to an extracellular epitope of the 55kd CD4 molecule present on a murine T cell subset (1,2). CD4 binds to MHC Class II expressed by antigen presenting cells, facilitating T Cell Receptor signaling. This clone has proven effective as a T cell depletion reagent(3).

NCBI Description

This gene encodes a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigenes and is also a receptor for the human immunodeficiency virus. This gene is expressed not only in T lymphocytes, but also in B cells, macrophages, and granulocytes. It is also expressed in specific regions of the brain. The protein functions to initiate or augment the early phase of T-cell activation, and may function as an important mediator of indirect neuronal damage in infectious and immune-mediated diseases of the central nervous system. Multiple alternatively spliced transcript variants encoding different isoforms have been identified in this gene. [provided by RefSeq, Aug 2010]
