Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for IL-33 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IL-33 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL-33 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL-33 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Cytokine that binds to and signals through the IL1RL1/ST2 receptor which in turn activates NF-kappa-B and MAPK signaling pathways in target cells. Involved in the maturation of Th2 cells inducing the secretion of T-helper type 2-associated cytokines. Also involved in activation of mast cells, basophils, eosinophils and natural killer cells. Acts as a chemoattractant for Th2 cells, and may function as an "alarmin", that amplifies immune responses during tissue injury
