Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for IGF-2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IGF-2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IGF-2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IGF-2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the insulin family of polypeptide growth factors, which are involved in development and growth. It is an imprinted gene, expressed only from the paternal allele, and epigenetic changes at this locus are associated with Wilms tumour, Beckwith-Wiedemann syndrome, rhabdomyosarcoma, and Silver-Russell syndrome. A read-through INS-IGF2 gene exists, whose 5' region overlaps the INS gene and the 3' region overlaps this gene.
