Related Product Information for IGF1R / CD221 recombinant protein
Background: The insulin-like growth factor-1 receptor (IGF1R) is a transmembrane tyrosine kinase involved in several biological processes including cell proliferation, differentiation, DNA repair, and cell survival. This a disulfide-linked heterotetrameric transmembrane protein consisting of two alpha and two beta subunits, and among which, the alpha subunit is extracellular while the beta subunit has an extracellular domain, a transmembrane domain and a cytoplasmic tyrosine kinase domain. IGF1R signalling pathway is activated in the mammalian nervous system from early developmental stages. Its major effect on developing neural cells is to promote their growth and survival. This pathway can integrate its action with signalling pathways of growth and morphogenetic factors that induce cell fate specification and selective expansion of specified neural cell subsets. Modulation of cell migration is another possible role that IGF1R activation may play in neurogenesis. In the mature brain, IGF-I binding sites have been found in different regions of the brain, and multiple reports confirmed a strong neuroprotective action of the IGF-IR against different pro-apoptotic insults. IGF1R is an important signaling molecule in cancer cells and plays an essential role in the establishment and maintenance of the transformed phenotype . Inhibition of IGF1R signaling thus appears to be a promising strategy to interfere with the growth and survival of cancer cells. IGF1R is frequently overexpressed by tumours, and mediates proliferation and apoptosis protection. IGF signalling also influences hypoxia signalling, protease secretion, tumour cell motility and adhesion, and thus can affect the propensity for invasion and metastasis. Therefore, the IGF1R is now an attractive anti-cancer treatment target.

Description: A DNA sequence encoding the extracellular domain (Met 1-Asn 932) of human IGF1 Receptor (NP_000866.1) precursor was expressed with a C-terminal polyhistidine tag.

NCBI Description

This receptor binds insulin-like growth factor with a high affinity. It has tyrosine kinase activity. The insulin-like growth factor I receptor plays a critical role in transformation events. Cleavage of the precursor generates alpha and beta subunits. It is highly overexpressed in most malignant tissues where it functions as an anti-apoptotic agent by enhancing cell survival. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, May 2014]

Uniprot Description

IGF1R: a receptor tyrosine kinase that binds insulin-like growth factor 1 (IGF1) with a high affinity and IGF2 with a lower affinity. Functions as an anti-apoptotic agent by enhancing cell survival. Can play a critical role in transformation events. Cleavage of the precursor generates alpha and beta subunits. Tetramer of 2 alpha and 2 beta chains linked by disulfide bonds. The alpha chains contribute to the formation of the ligand- binding domain, while the beta chain carries the kinase domain. Interacts with PIK3R1 and with the PTB/PID domains of IRS1 and SHC1 in vitro when autophosphorylated on tyrosine residues. Mutated in rare cases of pre- and post-natal growth retardation. One SNP associated with increased human longevity. Increased expression of IGF1R and other pathway members associated with progression and malignancy in a range of cancers. Inhibitors: AG1024, AEW541.

Product Notes

The IGF1R / CD221 igf1r (Catalog #AAA2546468) is a Recombinant Protein produced from Human Cells and is intended for research purposes only. The product is available for immediate purchase. The calculated molecular masses for alpha subunit, beta subunit and the single chain of the receptor are approximately 81 kDa, 23 kDa and 104 kDa, respectively. As a result of glycosylation, the recombinant single chain, alpha subunit and beta subunit have apparent molecular masses of approximately 150 kDa, 120 kDa and 48 kDa, respectively in SDS-PAGE under reducing conditions. Researchers should empirically determine the suitability of the IGF1R / CD221 igf1r for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IGF1R / CD221, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
