Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Target of rapamycin complex 2 subunit MAPKAP1 (MAPKAP1) ELISA Kit employs a two-site sandwich ELISA to quantitate MAPKAP1 in samples. An antibody specific for MAPKAP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MAPKAP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAPKAP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAPKAP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Target of rapamycin complex 2 subunit MAPKAP1 is a protein similar to the yeast SIN1 protein, a stress-activated protein kinase. Alternatively spliced transcript variants encoding distinct isoforms have been described. Alternate polyadenylation sites as well as alternate 3' UTRs have been identified for transcripts of this gene. The deduced 522-amino acid protein has a nuclear localization signal, 2 bipartite nuclear localization signals, a peroxisomal targeting signal, and a PEST motif for rapid protein degradation. Comparison of SIN1 with homologs from various species revealed a short, highly conserved region, designated Box1, located within a larger conserved domain, designated CRIM (conserved region in middle).

This gene encodes a protein that is highly similar to the yeast SIN1 protein, a stress-activated protein kinase. Alternatively spliced transcript variants encoding distinct isoforms have been described. Sin1: a member of a conserved family of proteins that have an essential roles in signal transduction from yeast to mammals. An essential component of the mTORC2 complex, which consists of mTOR, LST8, Sin1, Protor (PRR5), and RICTOR, the Rapamycin Insensitive Companion of mTOR. mTORC2 is activated by growth factors, but, in contrast to mTORC1, seems to be nutrient-insensitive. mTORC2 seems to function upstream of Rho GTPases to regulate the actin cytoskeleton, probably by activating one or more Rho-type guanine nucleotide exchange factors. mTORC2 promotes the serum-induced formation of stress-fibers or F-actin. mTORC2 regulates the phosphorylation of SGK1 S422 and PKCA S657. Enhances osmotic stress-induced phosphorylation of ATF2 and ATF2-mediated transcription. Contains a Ras-binding (RBD) and a pleckstrin homology (PH) domains. Plays an important role in the SAPK signaling pathway by binding directly to both ATF-2 and p38. A genetic knockout of Sin1 abolished Akt-S473 phosphorylation and disrupts the RICTOR-mTOR interaction, but Akt-Thr308 phosphorylation is maintained. Six alternatively-spliced isoforms of the human protein have been reported. All isoforms except 4 can be incorporated into the the mTORC2 complex.
