Intended Uses: This FCGR3B ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human FCGR3B. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: FCGR3B ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FCGR3B antibody and an FCGR3B-HRP conjugate. The assay sample and buffer are incubated together with FCGR3B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FCGR3B concentration since FCGR3B from samples and FCGR3B-HRP conjugate compete for the anti-FCGR3B antibody binding site. Since the number of sites is limited, as more sites are occupied by FCGR3B from the sample, fewer sites are left to bind FCGR3B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FCGR3B concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a low affinity receptor for the Fc region of gamma immunoglobulins (IgG). The encoded protein acts as a monomer and can bind either monomeric or aggregated IgG. This gene may function to capture immune complexes in the peripheral circulation. Several transcript variants encoding different isoforms have been found for this gene. A highly-similar gene encoding a related protein is also found on chromosome 1. [provided by RefSeq, Aug 2012]
