Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to Gastric Intrinsic Factor (GIF) has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled Gastric Intrinsic Factor (GIF) and unlabeled Gastric Intrinsic Factor (GIF) (Standards or samples) with the pre-coated antibody specific to Gastric Intrinsic Factor (GIF). After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of Gastric Intrinsic Factor (GIF) in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of Gastric Intrinsic Factor (GIF) in the sample.

NCBI Description

This gene is a member of the cobalamin transport protein family. It encodes a glycoprotein secreted by parietal cells of the gastric mucosa and is required for adequate absorption of vitamin B12. Vitamin B12 is necessary for erythrocyte maturation and mutations in this gene may lead to congenital pernicious anemia. [provided by RefSeq, Jul 2008]

GIF: Promotes absorption of the essential vitamin cobalamin (Cbl) in the ileum. After interaction with CUBN, the GIF-cobalamin complex is internalized via receptor-mediated endocytosis. Defects in GIF are the cause of hereditary intrinsic factor deficiency (IFD); also known as congenital pernicious anemia. IFD is an autosomal recessive disorder characterized by megaloblastic anemia. Belongs to the eukaryotic cobalamin transport proteins family. 2 isoforms of the human protein are produced by alternative splicing.
