Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. anti-Human GIF antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human GIF antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human GIF amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Human GIF can be calculated.

NCBI Description

This gene is a member of the cobalamin transport protein family. It encodes a glycoprotein secreted by parietal cells of the gastric mucosa and is required for adequate absorption of vitamin B12. Vitamin B12 is necessary for erythrocyte maturation and mutations in this gene may lead to congenital pernicious anemia. [provided by RefSeq, Jul 2008]

Defects in GIF are the cause of hereditary intrinsic factor deficiency (IFD); also known as congenital pernicious anemia. IFD is an autosomal recessive disorder characterized by megaloblastic anemia. Belongs to the eukaryotic cobalamin transport proteins family. 2 isoforms of the human protein are produced by alternative splicing.
