Principle of the Assay: The Human CD28 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CD28 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human CD28 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD28 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CD28 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD28 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: CD28 (Cluster of Differentiation 28) is one of the proteins expressed on T cells that provide co- stimulatory signals required for T cell activation and survival. T cell stimulation through CD28 in addition to the T-cell receptor (TCR) can provide a potent signal for the production of various interleukins (IL-6 in particular). The CD28 gene is mapped to 2q33-q34. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily.
