Principle of the Assay: The LumiAb Human GM-CSF Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human GM-CSF concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human GM-CSF while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human GM-CSF, also known as Granulocyte-Macrophage Colony-Stimulating Factor, is a 144 amino acid cytokine protein that stimulates the growth and differentiation of hematopoietic precursor cells from various lineages, including granulocytes, macrophages, eosinophils and erythrocytes. Encoded by the CSF2 gene at locus 5q31.1 of chromosome 5, this gene has been localized to a cluster of related genes at the same region, which is known to be associated with the interstitial deletions in the 5q-syndrome and acute myelogenous leukemia; other genes in the clutter include those encoding IL-4, IL-5 and IL-13. After initial synthesis, the protein is proteolytically cleaved to separate the 17 residue signal sequence from the 127 GM-CSF peptide, allowing for the maturation and proper folding of the actual cytokine. The active form of the protein is often found extracellularly as a homodimer. The cytokine is also used pharmaceutically in myeloid reconstitution following bone marrow transplant, bone marrow transplant engraftment failure or delay, mobilization and following transplantation of autologous peripheral blood progenitor cells, and following induction chemotherapy in older adults with acute myelogenous leukemia.

NCBI Description

This antimicrobial gene encodes a member of the CXC subfamily of chemokines. The encoded protein is a secreted growth factor that signals through the G-protein coupled receptor, CXC receptor 2. This protein plays a role in inflammation and as a chemoattractant for neutrophils. Aberrant expression of this protein is associated with the growth and progression of certain tumors. A naturally occurring processed form of this protein has increased chemotactic activity. Alternate splicing results in coding and non-coding variants of this gene. A pseudogene of this gene is found on chromosome 4. [provided by RefSeq, Sep 2014]
