Reconstitution
It is recommended that sterile water (400 uL) be added to the vial to prepare a stock solution of 0.25 mg/mL/.
Concentration is measured by UV-Vis.
Endotoxin
< 1.0 EU per ug of the protein as determined by the LAL method
Bio-Activity
Measured by its binding ability in a functional ELISA. Inmobilized human B7-H2 at 1 ug/mL/ (100 uL/ well.) can bind human ICOS with a linear range of 1.6-200- ng/mL.
Preparation and Storage
Store it under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted and be used as soon as possible. Avoid repeated freeze-thaw cycles.

Related Product Information for ICOS / AILIM / CD278 active protein
Background: Inducible costimulator (ICOS), also called AILIM (activiation-inducible lymphocyte immunomediatory molecule) is a cell-surface receptor, and belongs to the CD28 family of immune costimulatory receptors consisting of CD28, CTLA-4 and PD-1. The interaction of B7-H2/ICOS plays a critical role in Th cell differentiation, T-B cell interactions which is essential for germinal center formation, and humoral immune responses, and as well as the production of cytokine IL-4. In addition, ICOS is more potent in the induction of IL-10 production, a cytokine important for suppressive function of T regulatory cells. The B7-1/B7-2--CD28/CTLA-4 and ICOS-B7RP-1 pathway provides key second signals that can regulate the activation, inhibition and fine-tuning of T-lymphocyte responses. ICOS stimulates both Th1 and Th2 cytokine production but may have a preferential role in Th2 cell development. Moreover, The B7-1/B7-2-CD28/CTLA-4 and ICOS-B7RP-1 pathway has been suggested of being involved in the development of airway inflammation and airway hyperresponsiveness.

Description: A DNA sequence encoding the extracellular domain of human ICOS (NP_036224.1) (Met 1-Phe 141) was fused to the C-terminal polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

NCBI Description

The protein encoded by this gene belongs to the CD28 and CTLA-4 cell-surface receptor family. It forms homodimers and plays an important role in cell-cell signaling, immune responses, and regulation of cell proliferation. [provided by RefSeq, Jul 2008]

Uniprot Description

ICOS: Enhances all basic T-cell responses to a foreign antigen, namely proliferation, secretion of lymphokines, up- regulation of molecules that mediate cell-cell interaction, and effective help for antibody secretion by B-cells. Essential both for efficient interaction between T and B-cells and for normal antibody responses to T-cell dependent antigens. Does not up- regulate the production of interleukin-2, but superinduces the synthesis of interleukin-10. Prevents the apoptosis of pre- activated T-cells. Plays a critical role in CD40-mediated class switching of immunoglobin isotypes. Defects in ICOS are the cause of immunodeficiency common variable type 1 (CVID1). CVID1 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be lo. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The ICOS / AILIM / CD278 icos (Catalog #AAA2546316) is an Active Protein produced from Human Cells and is intended for research purposes only. The product is available for immediate purchase. The recombinant human ICOS/Fc is a disulfide-linked homodimeric protein. The reduced monomer consists of 368 amino acids and predicts a molecular mass of 41.6 kDa. The apparent molecular mass of rhICOS/Fc monomer is approximately 50 kDa due to glycosylation. Researchers should empirically determine the suitability of the ICOS / AILIM / CD278 icos for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ICOS / AILIM / CD278, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
