Human Hyaluronate binding protein ELISA Kit
Principle of the Assay: HABP ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-HABP antibody and an HABP-HRP conjugate. The assay sample and buffer are incubated together with HABP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HABP concentration since HABP from samples and HABP-HRP conjugate compete for the anti-HABP antibody binding site. Since the number of sites is limited, as more sites are occupied by HABP from the sample, fewer sites are left to bind HABP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HABP concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a secretory protein that contains a hyaluronan-binding domain, and thus is a member of the hyaluronan-binding protein family. The hyaluronan-binding domain is known to be involved in extracellular matrix stability and cell migration. This protein has been shown to form a stable complex with inter-alpha-inhibitor (I alpha I), and thus enhance the serine protease inhibitory activity of I alpha I, which is important in the protease network associated with inflammation. This gene can be induced by proinflammatory cytokines such as tumor necrosis factor alpha and interleukin-1. Enhanced levels of this protein are found in the synovial fluid of patients with osteoarthritis and rheumatoid arthritis.[provided by RefSeq, Dec 2010]
