Principle of the Assay: GAL10 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-GAL10 antibody and an GAL10-HRP conjugate. The assay sample and buffer are incubated together with GAL10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GAL10 concentration since GAL10 from samples and GAL10-HRP conjugate compete for the anti-GAL10 antibody binding site. Since the number of sites is limited, as more sites are occupied by GAL10 from the sample, fewer sites are left to bind GAL10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAL10 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the galectin family of carbohydrate binding proteins. Members of this protein family have an affinity for beta-galactosides. The encoded protein is characterized by an N-terminal proline-rich tandem repeat domain and a single C-terminal carbohydrate recognition domain. This protein can self-associate through the N-terminal domain allowing it to bind to multivalent saccharide ligands. This protein localizes to the extracellular matrix, the cytoplasm and the nucleus. This protein plays a role in numerous cellular functions including apoptosis, innate immunity, cell adhesion and T-cell regulation.
