Principle of the Assay: IL1D ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL1D antibody and an IL1D-HRP conjugate. The assay sample and buffer are incubated together with IL1D-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL1D concentration since IL1D from samples and IL1D-HRP conjugate compete for the anti-IL1D antibody binding site. Since the number of sites is limited, as more sites are occupied by IL1D from the sample, fewer sites are left to bind IL1D-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL1D concentration in each sample is interpolated from this standard curve.

IL1F5: Is a highly and a specific antagonist of the IL-1 receptor-related protein 2-mediated response to interleukin 1 family member 9 (IL1F9). Could constitute part of an independent signaling system analogous to interleukin-1 alpha (IL-1A), beta (IL-1B) receptor agonist and interleukin-1 receptor type I (IL- 1R1), that is present in epithelial barriers and takes part in local inflammatory response. Defects in IL36RN are the cause of psoriasis generalized pustular (PSORP). PSORP is a life-threatening disease defined by repeated flares of sudden onset consisting of diffuse erythematous skin eruption characterized by rapid coverage with pustules, high-grade fever, asthenia, marked leukocytosis, and elevated serum levels of C-reactive protein. Belongs to the IL-1 family.
